Retatrutide is Eli Lilly's LY3437943. A 39-residue peptide, built one amino acid at a time on a solid support. Where tirzepatide pulls on two incretin pathways, retatrutide pulls on three. The third one — glucagon — is what makes the molecule interesting. It is also what makes it incomplete: phase 3 readouts are still pending at the time of writing.
